  Oligonculeotide Synthesis


Oligonucleotides are short, single strand, nucleic acid sequences. DNA oligonucleotides have a phosphate backbone, a 2´-deoxyribose sugar and the 4 bases adenine, thymine, guanine and cytosine:

Chemically, the difference between DNA and RNA is in the sugar constituents. In RNA the sugar is a ribose, which means there is a hydroxyl (OH) function at the 2´ position of the sugar (as highlighted in red on the two bases in the diagram). Furthermore, RNA has uracil as a base instead of thymine. Modern parallel synthesis devices are used to produce oligos efficiently, in great quantities and with manifold modifications. Today’s larger synthesisers are capable of producing quantities of more than 500g of oligonucleotide per synthesis. We use both types of synthesiser at BioSpring. The possibility of producing oligos chemically has opened up a number of new areas of application in the last few years. One of the best known is molecular biology. Both simple primers for polymerase chain reactions (“PCR”) and complex colour-coded oligos for real-time PCR detection are used in this field of application. Oligos have also become indispensible tools in diagnostics. One need only consider the detection of viruses (e.g. HCV and HIV) and genetic diseases (e.g. illnesses caused by single nucleotide polymorphism or SNP). The use of oligonucleotides in chip technology has also become widely spread. The oligonucleotides are immobilised on a solid surface where they function as specific "receptors". The use of nucleic acids as therapeutic agents is becoming increasingly important, especially the employment of oligonucleotides as immunomodulators, expression blockers and simple peptide mimetics.
