We use the term chimaera to describe oligonucleotides composed of a combination of diverse components such as DNA and RNA or backbone modifications such as thioate/phosphate.

Chimaera oligonucleotides are a bit more complicated to order. Once you select Chimaera from the Type menu, your oligonucleotide sequence will be defined using three letters per base:

The first letters (d, r, q and o) stand for:

I. Desoxyribonucleotide; d
II. Ribonucleotide; r
III. 2’Amino-Ribonucleotide; q
IV. 2’Methoxy-Ribonucleotide; o

The second letter defines which base you would like, as usual.

The third letter determines which link should be used between two components (p or s). You have the choice between a phosphate (p) and a thiophosphate (s) modification:

I. Phosphate; p
II. Thioate; s

To repeat, when ordering chimaeras you always have to choose three letters to be able to define a position in your sequence. Naturally, you can also access the Help function to get more useful information on entering your sequence. Please note: At the 3’ end of your sequence, the last base is only defined using the first two letters. In this modus, you are only able to enter modifications on the end of your sequence. In this case, please enter the modification as follows:

I. 5’ modification; phosphate XXp or thioate XXs
II. 3’ modification; XX

 

To make it easier, some different information is needed for ordering RNAi oligonucleotides. We make a distinction between the RNA and the DNA part of the RNAi molecule. The RNA part is composed of 19-25 RNA single components (a, c, g and u) and is found on the 5’ end of the RNAi molecule. In contrast, the DNA sequence is composed of 0-4 single components (a, c, g and t) found on the 3’ end of the RNA sequence. Information entered into the DNA window is then automatically added to the 3’ end of the RNA sequence. When the completed sequence is put in the shopping cart, this section appears with the abbreviation d (e.g. dt). Special modifications are possible and can be marked using an X.
